Katie B will need one - although she was above the top seed in qualifying on the entry list, she was below 4th Q seed Muchova and 2nd Q seed Frech (it's not always safe just to look at who the top Q seed is since the rankings for entry and seeding aren't taken at the same dates), so if she'd got in as a direct acceptance, Muchova and Frech would have got in too and wouldn't be in the qualifying draw.

Both players you have mentioned are below Katie in the updated rankings though, Muchova way down now in fact.
Changes in rankings since the entry cut-off date affect seedings (so if Katie had played in qualifying, she'd have been the top Q seed) but I'm afraid they make no difference to who gets direct entry to the main draw - that is decided by the rankings at the entry cut-off date, i.e. those shown on the acceptance list on ITF.
So she'll be in the main draw (unless she has injured herself in the last couple of days - hopefully not!) but she will use up one of the four wild cards.

Yes, just possible (though unlikely) that Sam has one of the wildcards.
After all, she made the quarters here last year, trouncing Gabi T, 6-3 6-0, on the way.
That should count for something.....
It wasn't enough for her to get a wild card into Wimbledon qualifying last year, which was a lot more shocking than her not getting a main draw wild card here would be. Presumably, though, one of the three remaining (realistic) possibles is injured though, otherwise why wouldn't Sam be in the qualifying draw.
I am afraid that I would guess the missing ( at least ) one to be Sam since I doubt she would have got a WC here before some in the qualifying draw unless maybe some local reason ?
And yes, Sam's non Wimbledon qualifying WC last year was an absolutely unwarranted shocker.
So play was due to start at 11am. Even after warm-ups that mean play should hve started proper at 11.10. Nothing yet. What's the weather like down there?
